Cbf5 belongs to the TruB family of pseudouridine synthases and is a core component of the box H/ACA RNP complex in archaea and eukaryotes, working alongside Nop10, Gar1, and L7Ae to catalyze site-specific pseudouridylation (Ψ), guided by H/ACA RNAs. Cbf5 recognizes the ACA motif and lower stem of H/ACA RNAs, while L7Ae binds the K-turn motif. Cbf5 can also modify RNA independently of guide RNAs - two atypical H/ACA RNAs (sR563, sR109) were shown to be direct substrates of Cbf5, with pseudouridines (e.g., U7, U9, U5, U14) formed in their own 5′ guide regions, indicating Cbf5 acts directly on them. Was found to be essential in S. islandicus.
